The Mouse IGF Binding Protein 3 (Ms IGFBP-3) ELISA quantitates Ms IGFBP-3 in mouse serum, plasma, or cell culture medium. The assay will exclusively recognize both natural and recombinant Ms IGFBP-3. Principle of the method The Mouse IGFBP-3 solid-phase sandwich ELISA (enzyme-linked immunosorbent assay) is designed to measure the amount of the target bound between a matched antibody pair. A target-specific antibody has been pre-coated in the wells of the supplied microplate. Samples, standards, or controls are then added into these wells and bind to the immobilized (capture) antibody. The sandwich is formed by the addition of the second (detector) antibody, a substrate solution is added that reacts with the enzyme-antibody-target complex to produce measurable signal. The intensity of this signal is directly proportional to the concentration of target present in the original specimen. La protéine de liaison du facteur de croissance similaire à l’insuline 3 (IGFBP3) fait partie de la superfamille des protéines de liaison du facteur de croissance similaire à l’insuline (IGF), qui comprennent six protéines de liaison IGF à haute affinité (IGFBP) et au moins quatre protéines de liaison à faible affinité appelées protéines liées à l’IGFBP (IGFBPrP). Les membres de la famille IGFBP sont des protéines riches en cystéine avec des résidus de cystéine conservés regroupés dans les régions d’extrémité carboxylique et aminoterminale de la molécule. La séquence d’ADNc codant l’IGFBP3 humaine mature est fusionnée au peptide signal de CD33. Le résidu à 265 acides aminés de l’IGFBP3 mature recombinante humaine a une masse moléculaire calculée d’environ 29 kDa. En raison de la glycosylation, la protéine recombinante migre en 41 kDa en SDS-PAGE.
